The Effect of Overexpression of Foxp3 on Mice Tumorigenesis

Abstract


Aims: Study the effect of overexpression of transcription factor Foxp3 in lung cancer cell on tumorigenesis. Method: establish NCIH-1299 lung cancer cell strain with stable highly-expressed Foxp3 by using transfection method mediated by liposome as the positive cell of highly expressed Foxp3, and transfect NCIH-1299 cell by using pcDNA-3 empty carrier as negative control. Adopt ELISA method to detect the change of cytokine expression quantity such as IL-8, IL-10; establish transplantation tumor mouse model, observe the function of Foxp3 in tumorigenesis in mouse. Result: successfully established NCIH-1299 lung cancer cell strain with highly-expressed Foxp3; compared with negative control group, IL-8 the expression quantity decreases in cells of highly expressed Foxp3, which of IL-10 increases. Use transplantation tumor model mice to observe the difference, and find that NCIH - 1299 pcdna3 - hFoxp3 mice appear tumor after 18 days of inoculation, with a faster speed and larger tumor diameter, NCIH - 1299 groups of mice appear tumor after 25 days of inoculation , with a slower speed and smaller tumor diameter. Conclusion: high expression of Foxp3 promotes the secretion of IL - 10 lung cancer cell and inhibits the expression of IL - 8, promotes the formation and growth of tumor.
